Specificity Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Alkaline Phosphatase (E. coli). Cross-reactivity against Alkaline Phosphatase from other tissues and species may occur but have not been specifically determined. Some minimal cross-reactivity has been reported when lysates of cell extracts overproducing E. coli proteins are probed.

Application Details

Application Notes Assayed against 1ug of Alkaline Phosphatase (E. coli) in a standard ELISA using Peroxidase conjugated Affinity Purified Goat anti Rabbit IgG (H&L) and ABTS as a substrate for 30 minutes at room temperature. A working dilution of 1:25,000 to 1:125,000 of the reconstitution concentration is suggested for this product. A 1:10,000 dilution has been reported to be useful for the detection by immunoblot of an integral membrane protein from a gram-positive organism expressed as a LacZ/PhoA chimeric derivation of E. coli. Each laboratory should determine an optimum working titer for use in its particular application. Other applications have not been tested but use in such assays should not necessarily be excluded.

Buffer Lyophilized from 0.02M Potassium Phosphate, 0.15M Sodium chloride, pH 7.2. Preservative: 0.01% Sodium azide
Storage Store lyophilized product at 2-8°C. After reconstitution, this product is stable for several weeks at 2-8°C as an undiluted liquid. Prepare working dilution only prior to immediate use. For extended storage, aliquot and store at -20°C or below. Avoid multiple freeze/thaw cycles. Expiration date is one (1) year from date of reconstitution. : This product contains sodium azide, which has been classified as Xn (Harmful) in European Directive 67/548/EEC in the concentration range of 0.1-1.0%. When disposing of this reagent through lead or copper plumbing, flush with copious volumes of water to prevent azide build-up in drains.Reconstitute with 5ml of deionized water (or equivalent). Centrifuge product if not completely clear after standing at room temperature.
