Description The oncoprotein ErbB2/HER2 (human epidermal growth factor receptor 2), also known asNeu, is a 185 kDa transmembrane tyrosine kinase of cell surface growth factor receptorfamily. It is present in a wide variety cell types of normal human fetal and adult tissues andis frequently overexpressed in human carcinomas (e. g. in 20-30% cases of breast cancercells). Activation of ErbB2 triggers intracellular signalling events, which are essential forcell growth and differentiation. In the last 20 years ErbB2 antigen has become veryimportant marker and therapy target in patient care.
Specificity The polyclonal antibody reacts with ErbB2/HER2 oncogen, a 185 kDa transmembranetyrosine kinase presented in a wide variety cell types of human tissues. Species: Human. Others not tested.

Application Details

Application Notes Immunohistochemistry (paraffin sections): Recommended dilution: 10 μg/ml in PBS with 10% FCS and 0.09 % NaN3Staining technique: standard ABC technique (DAB+)Pretreatment: Fill the staining dishes with 10 mM citrate buffer pH 6.0 (pretreatmentsolution). Put the staining dishes with the pretreatment solution in the water bath and heaton 95-99C. Sink the deparaffin sections into the preheated pretreatment solution. Onceagain heat the water bath and pretreatment solution with sections on 95-99C - incubatefor 40 min in 95-99C. Remove the dishes with sections from the water bath and keepthem cool down for 20 min in RT. Rinse the sections 3 x 5 min in washing buffer (PBS). Incubation: overnight at 4CPositive tissue: breast carcinomaNote: Warning: The sections must not drain! ! . Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
