Description
HLA-G belongs to the HLA class I heavy chain paralogues (MHC Class Ib, nonclassical). It isa heterodimer consisting of a heavy chain (with a1, a2, a3 domains) and a light chain (beta2 microglobulin). Alternate splicing provides both soluble and membrane bound forms ofHLA G, with two soluble isoforms and four membrane bound isoforms. HLA G may be partof the fetal protection system and is expressed on fetal derived placental cells. It is expressed on the surface of trophoblast cells.
Specificity
ABIN238376 recognizes both membrane-bound and soluble forms of HLA-G (HLA-G1 andHLA-G5). This antibody blocks interaction of HLA-G with inhibitory receptors. Species Reactivity: Tested: Human. Does not reacts with Mouse and Rat.

Application Notes
Flow Cytometry. Positive control: JEG-3 human choriocarcinoma epithelial cell lineImmunohistochemistry (Frozen Sections): 10 µg/ml (20 min at 25°C). Positive tissue: extravillous cytotrophoblastELISA: Has been tested as the Capture antibody in a sandwich ELISA for analysis of humanHLA-G in combination with antibody W6/32. Recommended Dilution: 1 µg/mlPositive Control: JEG-3 human choriocarcinoma epithelial cell line. Functional Application: This antibody 87G blocks interaction of HLA-G with inhibitoryreceptors. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
