Description: Zeaxanthin Epoxidase (ZEP) catalyses the O2-/NAPDHdependent epoxidation ofzeaxanthin to violaxanthin via antheraxanthin at the stromal side of the thylakoidmembrane of green plants. ZEP also functions in the first step of the biosynthesisof the abiotic stress hormone abscisic acid (ABA).

Specificity: Predicted reactivity: dicots including: Brassica rapa, Vitis vinifera. Not reactive in: Spinacia oleracea.

10 ug of total leaf protein from (1) A.thaliana leafs isolated with PEB (see product AS08 300) and A.thaliana chloroplasts (2) and thylakoids (3) where separated on 4-12% NuPage (Invitrogen) LDS-PAGE and blotted 60 min (30V) to nitrocellulose. Filters where blocked 1h with 2% low-fat milk powder in TBS-T (0.1% TWEEN 20) and probed with AS08 289 (1:1000, 1h) and secondary anti-rabbit (1:20000, 1 h) antibody (HRP conjugated, Abcam) in TBS-T containing 2% low fat milk powder. Antibody incubations where followed by washings in TBS-T (15, +5, +5, +5 min). All steps were performed at RT with agitation. Signal was detected with standard ECL (Invitrogen) using a Fuji LAS-3000 CCD (150s, standard sensitivity).
