Description
|
SPHK1 catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate(SPP), a lipid mediator which has both intra- and extracellular functions. Intracellularly,SPP regulates proliferation and survival. Extracellularly it functions as a ligand forEDG1. SPHK1 also acts on D-erythro-sphingosine and to a lesser extent sphinganine,but not other lipids, such as D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine,diacylglycerol, ceramide, or phosphatidylinositol. Competitive inhibitors of SPHK blockformation of SPP and selectively inhibit cellular proliferation induced by a variety offactors, including platelet-derived growth factor and serum. SPHK1 has also been foundto mediate TRAF2 activation (Xia et al 2002). TRAF2 is a member of the TNF receptorassociated family and is required for TNF-alpha activation of MAPK8/JNK and NFkB.SPHK1 is widely expressed, with highest levels in adult liver, kidney, heart and skeletalmuscle.
