Alternative Name NANOG (NANOG Antibody Abstract)
Background Nanog is a newly identified homeodomain-bearing transcriptional factor. Nanogexpression is specific to early embryos and pluripotential stem cells including mouseand human embryonic stem (ES) and embryonic germ (EG) cells. It is a key moleculeinvolved in the signaling pathway for maintaining the capacity for self-renewal andpluripotency, bypassing regulation by the STAT3 pathway. Nanog mRNA is present inpluripotent mouse and human cell lines, and absent from differentiated cells.Nanog-deficient ES cells lose pluripotency and differentiate into extraembryonicendoderm lineage. Thus it is one of the molecular markers suitable for recognizing theundifferentiated state of stem cells in the mouse and human.NANOG is a new marker for testicular carcinoma in situ and germ cell tumors.Nanog is a 304 amino acid polypeptide which lacks a signal sequence for secretion. Alternate Names: anti-Nanog homeobox antibody, anti-ENK antibody, anti-Early Embryo specificexpression NK family gene antibody, anti-Embryonic stem cell specific homeoboxprotein (Nanog) antibody, anti-Homeobox transcription factor Nanog antibody.
Gene Symbol: NANOG
Gene ID 79923, 71950, 414065
NCBI Accession NP_079141
Application Notes Immunohistochemistry (PMID: 21678472). Approx 38kDa band observed in Human Ovary lysates (calculated MW of 34.6kDa according to NP_079141.1). Immunofluorescence: Parts of a keratinocyte-derived colony of induced pluripotent stem cells are stained for Nanog (green). This antibody does not work in PFA-fixed frozen embryos and ESC from Mouse.
Recommended dilutions: Immunocytochemistry/Immunofluorescence 1:10-1:500, Immunohistochemistry 1:10-1:500, Immunohistochemistry-Paraffin 1:10-1:500, Peptide ELISA detection limit 1:64000, Western Blot 0.03-0.1 µg/mL
Restrictions For Research Use only
Concentration 0.5 mg/mL
Buffer Tris saline, pH 7.3 with 0.5 % bovine serum albumin, Sodium Azide
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Avoid freeze-thaw cycles
Storage -20 °C
Storage Comment -20 °C.
