Description Nerve growth factor (NGF) influences the survival and differentiation of a variety ofneuronal and nonneuronal cells. The actions of this polypeptide result from binding tospecific cell surface receptors which are present as both high and low affinity sites. Although both types of receptor bind NGF only the high affinity form is capable of signaltransduction. Cross-linking studies with 125I-NGF have shown that the high affinityreceptor of rat PC12 cells forms an Mr 158,000 complex with NGF while the low affinityreceptor forms an Mr 100,000 complex. Full length cDNA clones for both the human and ratNGF receptor genes have been produced and sequenced, revealing greater than 90%homology between the two species. Recently, it has been shown that cells which do notnormally respond to NGF only express low affinity receptors from transfected NGF genesbut cells which normally are NGF-responsive express high affinity receptors from the sameNGF sequences. This supports the hypothesis that the high affinity receptors have a uniquecomponent in addition to the subunit common to the low affinity receptors.
Specificity Recognizes human, bovine and viper NGF. The epitope has been localized to aa 58-68 of hNGF. Species: Human, Mouse, Bovine and Viper. Others not tested.

Application Details

Application Notes ELISA: This antibody has been used as a coating antibody at 10 ug/ml in a two site ELISA withbiotin conjugated NGF antibody (Cat. No. ABIN264352) for mouse NGF. Typically formouse NGF, the ELISA has a working range of 0 -15 ng/ml and a sensitivity of 15 pg/ml withthe appropriate standard. Recommended sample volumes are 25-200 l. Recovery ofhuman NGF in human plasma is typically > 90% in the ELISA. The use of Tris buffer, pH 7, isrecommended. (Kenney, JS, Masada, MP, and Allison AC, Laboratory Methods inImmunology, Vol. I, 231, 1990). Neutralization: This antibody has been used to neutralize > 90% human NGF bioactivity onPC12 cells at a 200-fold molar excess. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
