Alternative Name CD184 / CXCR4 (CXCR4 Antibody Abstract)
Background A receptor for the C-X-C chemokine CXCL12/SDF-1, CXCR4 transduces a signal by increasing the intracellular calcium ions level. It is involved in haematopoiesis and in cardiac ventricular septum formation. Also plays an essential role in vascularization of the gastrointestinal tract, probably by regulating vascular branching and/or remodelling processes in endothelial cells. Could be involved in cerebellar development. In the CNS, could mediate hippocampal-neuron survival. Acts as a coreceptor (CD4 being the primary receptor) for HIV-1 X4 isolates and as a primary receptor for some HIV-2 isolates. Promotes Env-mediated fusion of the virus. Clone 12G5 has been shown to block infection by certain isolates of HIV-1 and HIV-2 and is specific for CXCR4 and does not react with IL8R-A, CCR1, CCR2b, CCR3, CCR4, and CCR5. Application Notes Flow CytometryImmunofluorescence. Immunocytochemistry. Immunohistochemistry on Frozen Sections. Immunohistochemistry on Paraffin Sections ( 10 μg/mL). Immunohistochemical analyses of the CXCR4 receptor in Human cell lines and tissues. Fixation with methanol and/or acetone was shown to destroy the epitope of the 12G5 mAb.12G5 can be used to block SDF-1-induced chemotaxis and increases of intracellularcalcium.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.
