Background Synapse-associated protein (SAP) 97 is a 97 kDa membraneassociated synapseprotein with extensive sequence homology to the PSD 95 family of proteins that facilitateion channel clustering at synaptic terminal. SAP97 consists of three 90 amino acid PDZdomains at its amino terminus, followed by a Src homology (SH) 3 domain, and a regionwith homology to guanylate kinases (GK). In addition to the CNS, SAP97 is alsodetected at the basal lateral membrane between a variety of epithelial cells. SAP97 isrecruited to the submembranous cortical cytoskeleton at cell-cell contact sites as part ofthe E-cadherin complex. Co-expression of SAP97 and K+ channel results in thecolocalization of these proteins in large round intracellular aggregates. SAP 97 has beenshown to have functional homology to Drosophila tumor suppressor protein (DlgA), aPSD 95 related protein involved in maintaining integrity of epithelial and neuromuscularjunctions.
