Background FOXP3 is a member of the forkhead/winged-helix family of transcriptional regulators andis highly conserved across mammals. FOXP3 is essential for normal immunehomeostasis. FOXP3 is stably and constitutively expressed at a high level in CD25 + CD4 positiveregulatory T cells, a low level in CD4-positive/CD25-negative cells, and is absent inCD4-negative/CD8-positive T cells. FOXP3 may be a master regulatory gene and amore specific marker of regulatory T cells than other T cells. Alternate Names: anti-AIID antibody, anti-DIETER antibody, anti-Forkhead box P3 antibody, anti-foxp3antibody, anti-IPEX antibody, anti-JM2 antibody, anti-PIDX antibody, anti-SCURFINantibody, anti-XPID antibody.

Application Notes This FOXP3 antibody is useful for Western blot, where a band can be seen at ~47.2 kDa representing FOXP3. This antibody only detects transfected samples. It has not been shown to work on endogenous samples via Western blot. Immunohistochemistry-Paraffin was reported in scientific literature.
Recommended dilutions: Immunohistochemistry-Paraffin, Western Blot 1:1000-1:5000
Protocol Protocol specific for FOXP3 Antibody IHC Protocol for NB600-246Control Tissues: Thymus from healthy (C57BL6/6) and Influenza infected mice.-Cut 5 um sections in a cryostat and fix them in cold acetone for 10 mins.-Dry the slides completely at room temp for 30 mins.-Permeabilize tissues by immersing them in 0.1 % saponin, 3 times (5 mins each time).-Block unspecific binding to the slides using 5 % normal donkey serum, 5 % normal rat serum and Fc block (1:500). Everything has to be dissolved in 0.1 % saponin.-Incubate slides in a wet chamber at RT for 30 mins.-Dip off the blocking solution from the slides and add streptavidin (0.1mg/ml) for 15 mins at RT.-Wash one time with 0.1 % saponin.-Add similar volume of biotin (0.
