Background NFkB is a transcription regulator that is activated by various intra and extra cellularstimuli such as cytokines, oxidant free radicals, ultraviolet irradiation, and bacterial orviral products. NFkB is a family of transcription factors that consists of homo andheterodimers of NFkB1/p50 and RelA/p65 subunits, and controls a variety of cellularevents including development and immune responses. All members share a conservedamino terminus domain that includes dimerization, nuclear localization, and DNAbinding regions, and a carboxy terminal transactivation domain. Serines 529 and 536 inthe transactivation domain of RelA/p65 are phosphorylated in response to severalstimuli including phorbol ester, IL1 alpha and TNF alpha as mediated by IkB kinase andp38 MAPK. Serine 529 is located in a negatively charged region (amino acids 422-540)that is phosphorylated in response to phorbol myristate acetate plus calcium ionophoreactivation. Phosphorylation of serines 529 and 536 is critical for RelA/p65 transcriptionalactivity. Activated NFkB translocates into the nucleus and stimulates the expression ofgenes involved in a wide variety of biological functions. Inappropriate activation of NFkBhas been associated with a number of inflammatory diseases while persistent inhibitionof NFkB leads to inappropriate immune cell development or delayed cell growth. Application Notes This product was assayed by immunoblot and found to be reactive against Human NFkB p50 at a dilution of 1:1000 followed by reaction with Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] Anti- Human NFkB p50 is suitable for the detection by immunoblot of Human NFkB p50 and its precursor protein p105. No reaction was observed against the analogous mouse protein. This product was also tested in a gel supershift assay and found to be reactive against p50:p50 homodimers and p:50:p65 heterodimers using 0.5 - 1.0 ul per assay.
