Background CRK7 is a CDC2/CDK -type protein kinase. Also known as CRKRS, the protein has anarginine/serine-rich (RS) domain that is found predominantly in splicing factors, andextensive proline-rich regions that match the consensus for SH3 and WW domainbinding sites. Anti-CrkRS immunoprecipitates have been shown to phosphorylate theC-terminal domain of RNA polymerase II in vitro. CRK7 has been shown to beexpressed ubiquitously. ESTs have been isolated from several tissue libraries, includingnormal lymph and cancerous bone. Alternate Names: Anti-Cell division cycle 2 related protein kinase 7 antibody, Anti-CRK7 antibody, Anti-EC2.7.11.22 antibody, Anti-Cdc2 related kinase arginine/serine rich antibody.

Protocol Protocol specific for CrkRS Antibody (NLS7350)Immunohistochemistry
1. Prepare tissue with formalin fixation and by embedding it in paraffin wax.
. Make 4 um sections and place on pre-cleaned and charged microscope slides.
. Heat in a tissue-drying oven for 45 minutes at 60 degrees Celcius.
. Deparaffinize the tissues by wash drying the slides in 3 changes of xylene for 5 minutes each at RT.
. Rehydrate the tissues by washing the slides in 3 changes of 100 % alcohol for 3 minutes each at RT.
. Wash the slides in 2 changes of 95 % alcohol for 3 minutes each at RT.
. Wash the slides in 1 change of 80 % alcohol for 3 minutes at RT.
. Rinse the slides in gentle running distilled water for 5 minutes at RT.
. Perform antigen retrieval by steaming the slides in 0.01M sodium citrate buffer (pH 6.
0) at 99-100 degrees Celciusfor 20 minutes.
. Remove the slides from the heat and let stand in buffer at RT for 20 minutes.
. Rinse the slides in 1X TBS-T for 1 minute at RT.**Do not allow the tissues to dry at any time during the staining procedure**
. Begin the immunostaining by applying a universal protein block for 20 minutes at RT.
. Drain protein block from the slides and apply the diluted primary antibody for 45 minutes at RT.
. Rinse the slide in 1X TBS-T for 1 minute at RT.
. Apply a biotinylated anti-rabbit IgG (H+L) secondary for 30 minutes at RT.
. Rinse the slide in 1X TBS-T for 1 minute at RT.
. Apply an alkaline phosphatase steptavidin for 30 minutes at RT.
. Rinse the slide in 1X TBS-T for 1 minute at RT.
. Apply an alkaline phosphatase chromagen substrate for 30 minutes at RT.
. Rinse the slide in distilled water for 1 minute at RT.**This method should only be used if the chromagen substrate is alcohol insoluble (ie: Vector Red, DAB)**
. Dehydrate the tissue by washing the slides in 2 changes of 80 % alcohol for 1 minute each at RT.
. Wash the slides in 2 changes of 95 % alcohol for 1 minute each at RT.
. Wash the slides in 3 changes of 100 % alcohol for 1 minute each at RT.
. Wash the slides in 3 changes of xylene for 1 minute each at RT.
. Apply cover slip.
