Background Members of the glial cell line derived neurotrophic factor (GDNF) family, including GDNFand neurturin (NTN), play key roles in the control of vertebrate neuron survival anddifferentiation. Physiological responses to NTN require the presence of a novelglycosylphosphadidylinositol linked protein NTNRa, which is a cell surface receptor forNTN. The cDNAs encoding NTNRa from human, rat, chicken, and mouse have beencloned recently. NTNRa was also termed GDNFRb, Ret ligand 2 (RETL2) or TGFbrelated neurotrophic factor receptor 2 (TrnR2) and nominated as GFRa2 recently.GFRa2 binds NTN and mediates activation of RET receptor tyrosine kinase by bothNTN and GDNF. Thus, NTN, GFRa2, and the Ret PTK form a complex to transduceNTN signal and to mediate NTN function. Application Notes Immunohistochemistry-P and Western Blot A dilution of 1:500 to 1:1000 is recommended for both applications. This antibody stains beautifully the large neurons in the rat DRG. This antibody also is ideal for WB analysis of rat DRG and brain homogenate. The optimal dilution should be determined by the end user, but a diution of 1 in 500 is recommended for initial testing.
