Immunogen A synthetic peptide (C-KNSASEE DHLPLQVLQSP) as part of rat VRL-1 protein (aa:744-761) conjugated to KLH.
Specificity Immunohistochemical analysis in rat dorsal root ganglia and spinal cord indicates a highlevel of specificity for this antiserum. Specificity was also shown by Western blot. Species Reactivity: This antibody is known to react with rat and human VRL-1. Localization: Accession Number: TRPV2_RAT.
Alternative Name TRPV2 / Vanilloid receptor-like protein 1
Background Caterina et al has identified a structurally related receptor, VRL-1, that does not respondto capsaicin, acid or moderate heat. Instead, VRL-1 is activated by high temperatures, with a threshold of approximately 52 degrees C. Within sensory ganglia, VRL-1 is mostprominently expressed by a subset of medium- to large-diameter neurons, making it acandidate receptor for transducing high-threshold heat responses in this class of cells.VRL-1 transcripts are not restricted to the sensory nervous system, indicating that thischannel may be activated by stimuli other than heat. VRL-1 is one of the six members ofthe vanilloid-like TRP-channel family which is now known as TRPV family. FUNCTION:Calcium-permeable, non-selective cation channel with an outward rectification. Seemsto be regulated, at least in part, by growth factors, like IGF1, PDGF and morphogeneticneuropeptide/head activator. May transduce physical stimuli in mast cells. Activated bytemperatures higher than 52 degrees Celsius, is not activated by vanilloids and acidicpH. SUBUNIT: Homotetramer (Probable). Interacts with a cAMP-dependent proteinkinase type II regulatory subunit (PRKAR2A or PRKAR2B) and ACBD3. Interacts withRGA. SUBCELLULAR LOCATION: Cell membrane, multi-pass membrane protein.Cytoplasm. Translocates from the cytoplasm to the plasma membrane upon ligandstimulation. TISSUE SPECIFICITY: Ubiquitously expressed. Expressed in dorsal rootganglia, trigeminal ganglia, spinal chord (Lissauer's tract, dorsal horn and dorsalcolumns) (at protein level). PTM: N-glycosylated. PTM: Phosphorylated by PKA.SIMILARITY: Belongs to the transient receptor family. TrpV subfamily. SIMILARITY:Contains 3 ANK repeats. Alternate Names: anti-Transient receptor potential cation channel subfamily V member 2 antibody,anti-TrpV2 antibody, anti-osm-9-like TRP channel 2 antibody, anti-OTRPC2 antibody,anti-Vanilloid receptor-like protein 1 antibody, anti-Stretch-activated channel 2B antibody. Related Diseases: TRP channels
Gene ID 51393, 29465
Application Notes IHC, Immunofluorescence, Western blot. Recommended to be used at a dilution of 1:1000 to 1: 2000 for immunohistochemistry and Western blot, for Immunofluorescence ata dilution of 1:50 to 1: 200 in free-floating sections or paraffin embedded sections. Theoptimal working dilution should be determined by the end user.
