Background Caterina et al has identified a structurally related receptor, VRL-1, that does not respondto capsaicin, acid or moderate heat. Instead, VRL-1 is activated by high temperatures,with a threshold of approximately 52 degrees C. Within sensory ganglia, VRL-1 is mostprominently expressed by a subset of medium- to large-diameter neurons, making it acandidate receptor for transducing high-threshold heat responses in this class of cells.VRL-1 transcripts are not restricted to the sensory nervous system, indicating that thischannel may be activated by stimuli other than heat. VRL-1 is one of the six members ofthe vanilloid-like TRP-channel family which is now known as TRPV family. FUNCTION:Calcium-permeable, non-selective cation channel with an outward rectification. Seemsto be regulated, at least in part, by growth factors, like IGF1, PDGF and morphogeneticneuropeptide/head activator. May transduce physical stimuli in mast cells. Activated bytemperatures higher than 52 degrees Celsius, is not activated by vanilloids and acidicpH. SUBUNIT: Homotetramer (Probable). Interacts with a cAMP-dependent proteinkinase type II regulatory subunit (PRKAR2A or PRKAR2B) and ACBD3. Interacts withRGA. SUBCELLULAR LOCATION: Cell membrane, multi-pass membrane protein.Cytoplasm. Translocates from the cytoplasm to the plasma membrane upon ligandstimulation. TISSUE SPECIFICITY: Ubiquitously expressed.
