Description P27 associates with cyclin and cyclin-dependent kinase complexes to inhibit their kinaseactivity and contribute to the control of cell proliferation. p27 is phosphorylated on manysites, including threonine 187, in vivo, with the predominant phosphorylation site beingserine 10. The extent of serine 10 phosphorylation by proline-directed kinase is markedlyincreased in cells in the G(0)-G(1) phase of the cell cycle compared to cells in the S or Mphase. p27 concentration is regulated predominantly by post-translational mechanisms. p27 is degraded by both the ubiquitin-proteasome pathway and ubiquitinindependentproteolysis. Regulation of ubiquitin-mediated proteolysis is often achieved throughubiquitination of the targeted phosphorylated protein, which renders it more susceptibleto degradation.
Specificity The p27 kip1 functions as a Negative regulator of G1 mouse expression and has beenproposed to function as a possible mediator of TGFb induced G1 arrest. Species: Human, Mouse, Rat, Monkey and Canine (Dog). Others not tested. Add. Information: Predicted Molecular Weight: 22. 1 kDa
