    Specificity
    This antibody recognises the CD103 cell surface antigen, also known as the alphaE integrin. This reacts with various subsets of intra-epithelial lymphocytes:- 40 % of lamina propria T-cells, 30 % of mesenteric lymphoblasts and some lymphocytes from other mucosa. It recognises less than 0.1 % bone marrow cells and less than 0.7 % PBL.

    Human Intestinal lymphocyte. Spleen cells from immunised BALB/c mice were fused with cells of the NS1 mouse myeloma cell line.

  • Application Notes
    Flow Cytometry: Use 10 μL of 1/50-1/100 diluted antibody to label 5 x 10e5 cells in 100 μL. Immunoprecipitation. Immunohistochemistry on Frozen Sections (1/50-1/100). Recommended positive control tissue: intestine.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background
    CD103/Integrin alpha E is a new member of the integrin series of adhesion molecules containing the beta7 subunit. This antigen defines a developmentally important subset of T cells, namely mucosal T cells including all IEL (intraepithelial lymphocytes) and ~20 % of lamina propria T cells. Expression of CD103 is more restricted outside these mucosal organs, appearing at lower levels on T cell subsets of the lymph node, dendritic epidermis and periphery. In non-epithelial CD103+ T cells there is a bias toward expression on CD8+ cells. Integrin alpha E is a heterodimer comprising a 150 kDa alpha chain and a 120 kDa beta chain. Integrin alpha E is expressed by approximately 1 % of peripheral blood lymphocytes, activated T lymphocytes and by hairy cell leukaemia cells.
