Immunogen: A synthetic peptide from human cysteine protease ATG4B (autophagin-1, APG4B, AUTL1) conjugated to an immunogenic carrier protein has been used as the immunogen. The antigen is homologous in mouse.

Description: Autophagy is the process by which endogenous proteins and damaged organelles are destroyed intracellularly.autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels ofautophagy have been described in some malignant tumors, and a role forautophagy in controlling the unregulated cell growth linked to cancer has been proposed. FUNCTION: Cysteine protease required forautophagy, which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes. Application Notes: IHC, WB. Use at a dilution of 1 : 200 to 1 : 2000. The optimal dilution should be determined by the end user. Not yet tested in other applications.

Purity: whole serum

Storage: Maintain the lyophilised/reconstituted antibodies frozen at -20°C for long term storage and refrigerated at 2-8°C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

WB on human neutrophil lysate using Rabbit antibody to human, mouse cysteine protease ATG4B (autophagin-1, APG4B, AUTL1): whole serum (ABIN350272) at a dilution of 1 : 100. Pre-absorption of the antibody with the immunising peptide completely abolishes the detected band.
