Immunogen A synthetic peptide from the c-terminal region of rat Orai1 (TMEM142a) conjugated to an immunogenic carrier protein was used as the antigen. The antigen is homologous in mouse and human.

Background Function: Ca2+ release-activated Ca2+ (CRAC) channel subunit which mediates Ca2+ influx following depletion of intracellular Ca2+ stores and channel activation by the Ca2+ sensor, STIM1. CRAC channels are the main pathway for Ca2+ influx in T-cells and promote the immune response to pathogens by activating the transcription factor NFAT. Defects in ORAI1 are a cause of severe combined immunodeficiency with CRAC channel dysfunction (CRAC-SCID). SCID refers to a genetically and clinically heterogeneous group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels. Patients with SCID present in infancy with recurrent, persistent infections by opportunistic organisms. The common characteristic of all types of SCID is absence of T-cell-mediated cellular immunity due to a defect in T-cell development.
