Description
Overgrowth of epithelial cells by fibroblasts has long been an obstacle to cultivation ofnormal and abnormal epithelial cells. In addition, apart from the functional significance ofcells characteristics, the characterization of cell lines is important for correlation ofcultures with tissue origin, identification of the lineage to which the cells belong or theprecursor status of the cells. Monoclonal antibodies to fibroblast surface protein shouldprovide a powerful tool for control of fibroblast growth in a variety of human cell culturesystems, such as thymic epithelial (TE) cell cultures allowing the growth of highly enrichedpopulations of the TE cells. It is also useful for Flow cytometry, Immunohistochemistry andImmunoblotting.

Specificity
This antibody reacts with proteins of 43 kDa and 72-80 kDa molecular weight. It reacts witha Surface Protein on Fibroblasts (synovial, foreskin and thymic), fibroblast cell lines, tissuemacrophages, and 95% of peripheral blood monocytes. It does not stain lymphocytes orhuman epithelial cells but does stain malignant fibrosarcoma tissue. Cellular Localization: Membrane. Species: Human. Others not tested.

Application Notes
Western Blot. Immunohistochemistry on Paraffin Sections (Pretreatment of formalin fixed paraffinembedded tissue sections has not been characterized). This product is intended for qualitative Immunohistochemistry with normal and neoplasticformalin-fixed, paraffin-embedded tissue sections, to be viewed by light microscopy. Clinical interpretation of staining results should be accompanied by histological studieswith proper controls. This antibody may be diluted to a titer of 1/25-1/50 in an ABC method for 30 min. at RT. Positive Control: Thymus. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
