Description: Long chain acyl-CoA synthetase (LACS), or long chain fatty acid-CoA ligases (FACLs),convert free long chain fatty acids into fatty acyl-CoA esters, which are key intermediates inthe synthesis of complex lipids. The deduced 683-amino acid FACL5 protein sharesapproximately 80% amino acid identity with the rat sequence. Northern blot analysisdetected major FACL5 transcripts of 2. 5 and 3. 7 kb in a many tissues, with highestexpression in uterus and spleen. Markedly increased levels of FACL5 transcripts aredetected in a glioma line and in primary gliomas of grade IV malignancy, Cultured gliomacells infected with an adenovirus encoding FACL5 displayed induced cell growth onexposure to palmitate. These results suggest a novel fatty acid-induced glioma cell growthmediated by FACL5.
