Description Actins are highly conserved proteins that are involved in various types of cell motility andare ubiquitously expressed in all eukaryotic cells. In vertebrates 3 main groups of actinisoforms, alpha, beta and gamma have been identified. The alpha actins are found inmuscle tissues and are a major constituent of the contractile apparatus. The beta andgamma actins coexist in most cell types as components of the cytoskeleton and asmediators of internal cell motility. Actins are highly conserved proteins expressed in all eucaryotic cells. Actin filaments formpart of the cytoskeleton and play essential roles in regulating cell shape and movement. Six distinct actin isotypes have been identified in mammalian cells. Each is encoded by aseparated gene and is expressed in a developmentally regulated and tissue-specificmanner, alpha and beta cytoplasmic actins are expressed in a wide variety of cells,whereas, expression of alpha skeletal, alpha cardiac, alpha vascular, and gamma entericactins are more restricted to specialized muscle cell type. Smooth muscle alpha actin is offurther interest because it is one of a few genes whose expression is relatively restricted tovascular smooth muscle cells. Further more, expression of smooth muscle alpha actin isregulated by hormones, cell proliferation , and altered by pathological conditions includingoncogenic transformation and atherosclerosis.
Specificity This antibody stains smooth muscle cells in vessel walls, gut wall, and myometrium. Myoepithelial cells in breast and salivary gland are also stained. It reacts with tumorsarising from smooth muscles and myoepithelial cells. Actin from fibroblasts (beta- andgamma-cytoplasmic), striated muscle (alpha-sarcomeric), and myocardium(alpha-myocardial) are not labeled. Application Notes Immunohistochemistry on Paraffin Sections: 1/100 for 10 min at RT. No special pretreatment is required for the immunohistochemical staining offormalin/paraffin tissues. Positiv Control: Smooth muscle or leiomyosarcoma. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
