Description
|
Securin is an anaphase inhibitor and plays an important role in DNA damage and spindlecheck point pathways. It inhibits sister chromatid separation by binding and inhibiting Esp1, a cystein protease that causes cleavage of the cohesin Scc 1 that binds the sisterchromatids together. It is responsible for targeting of Esp 1 to the nucleus and it’s bindingto the spindle. Degradation of Securin occurs shortly before anaphase, which liberates Esp1 and is a pre-requisite for anaphase entry. It is targeted for degradation by ubiquitinationmediated bycyclosome / anaphase promoting complex (APC) functioning as a ubiquitinligase. In response to DNA damage Chk 1 phosphorylates Securin to stabilize it against theAPC mediated destruction, hence preventing the entry of such a cell into anaphase. Securin is highly expressed in various human tumors, including pituitary, adrenal, kidney,liver, and ovarian tumors, and in cell lines derived from pituitary tumors, breast tumors,endometrial tumors, and ovarian tumors.

Immunohistochemistry on Paraffin Sections: 1/100 for 30min at RT, Boil tissue section in10mM citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min. Positive Control: Rectal carcinomaOther applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
