  • Application Notes
    Flow Cytometry: 10-20 μg/mL (final concentration). Positive Control: Peripheral blood Lymphocyte cells. Detailed procedure is provided in Protocols.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.
    Protocol
    Flow Cytometric analysis for floating cellsProtocol 1. We use Fisher tubes or equivalents as reaction tubes for all steps described below. 1) Wash the cells 3 times with washing buffer [PBS containing 2% fetal calf serum (FCS) and0. 1% NaN3]. 2) Add 10 μL of normal goat serum to the cell pellet after tapping. Mix well, and incubatefor 5 minutes at room temperature (20~25°C). 3) Add 30 µL of the FITC labeled CD161 monoclonal antibody (10-20 μg/mL) diluted with thewashing buffer. Mix well, and incubate for 30 minutes at room temperature (20~25°C). 4) Add 1 mL of the washing buffer followed by centrifugation at 500 x g for 1 minute at roomtemperature (20~25°C). Remove supernatant by careful aspiration. 5) Resuspend the cells with 500 μL of the washing buffer and analyze by a flow cytometer. Protocol 2We usually use Fisher tubes or equivalents as reaction tubes for all steps described below. 1) Wash the cells 3 times with washing buffer [PBS containing 2% fetal calf serum (FCS) and0. 1% NaN3]. 2) Resuspend the cells with PBS containing 25% normal goat serum and 0. 1% NaN3(5x10e6 cells/mL). 3) Add 20 µL of the non diluted FITC labeled CD161 monoclonal antibody into each tube. 4) Add 50 µL of the cell suspension into each tube. Mix well, and incubate for 30 minutesat room temperature (20~25°C). 5) Add 1 mL of the washing buffer followed by centrifugation at 500 x g for 1 minute at roomtemperature (20~25°C). Remove supernatant by careful aspiration. 6) Resuspend the cells with 500 µL of the washing buffer and analyze by a flow cytometer. Flow Cytometric analysis for whole blood cellsWe usually use Falcon tubes or equivalents as reaction tubes for all steps described below. 1) Add 20 μL of the non diluted FITC labeled CD161 monoclonal antibody into each tube. 2) Add 50 μL of whole blood into each tube. Mix well, and incubate for 30 minutes at roomtemperature (20~25°C). 3) Lyse with OptiLyse C (for analysis on Beckman Coulter instruments) or OptiLyse B (foranalysis on BD instruments), using the procedure recommended in the respective packageinserts. 4) Resuspend the cells with 500 μL of the washing buffer [PBS containing 2% fetal calfserum (FCS) and 0. 1% NaN3] and analyze by a flow cytometer.     Background
    CD161, also known as NKR-P1, is a receptor originally identified on the surface of natural killer cells that is also present on some subpopulations of CD8+T cells. CD161 is often used as an NK cell marker since it is present on both immature and mature NK cells. CD161 appears to mediate NK cell activation and is up-regulated by IL-12. CD161 is expressed as a disulfide homodimer of ~60 kDa while the ligand for CD161 appears to be a carbohydrate structure that includes GalNAc and GlcNAc structures.
