Description
|
Of the aldehydes which originate from the peroxidation of cellular membrane lipids,4-Hydroxy-2-Nonenal (HNE) is the major aldehyde generated by free radical attack on w-6polyunsaturated fatty acids. HNE is largely responsible for pathogenesis during oxidativestress. HNE is highly reactive towards free sulfydryl groups of proteins producing thioetheradducts that further undergo cyclization to form hemiacetals. HNE also reacts withhistidine and lysine residues of proteins to form stable Michael addition-type adducts. HNE induces heat shock protein, inhibits cellular proliferation and is highly toxic to cells. Itexhibits genotoxic and mutagenic effects as well. Elevated levels of HNE modified nigralneurons have also been observed in Parkinson disease.

Specificity
|
This antibody has a much higher affinity for the 4-Hydroxy-2-Noneal (4-HNE) Histidineadduct than the 4-HNE Lysine or 4-HNE Cysteine adducts. It shows almost negligible reactivity with proteins that were treated with other Aldehydes,such as 2-Nonenal, 2-Hexenal, 1-Hexanal, 4-Hydroxy-2-Hexenal, Formaldehyde orGlutaraldehyde.
