Description
|
Cyclic AMP-dependent phosphodiesterase type D (PDE4D) family is comprise of 5 variants(PDE4D1, D2, D3, D4, D6, D7, D8 and D9, (1, 2). One or more PDE4D subtype-variants areubiquitously present in all mammalian cells. In CNS all five PDE4D subtype-variants areexpressed in varying ratios and their activity is regulated in tandem with GPCRs stimulationand selective phosphorylation by PKA and other kinases. Peripheral tissues also exhibitdifferential expression of PDE4D variants. PDE4D1/D2 mRNA levels rise in response to anincrease in cAMP (3). Short-term regulation of PDE4D variants involved PKA, MAP kinasesand Erk2 phosphorylation that results in rapid change in their enzymatic activities (2, 4). Other regulatory mechanism involved protein-protein interactions with cytoskeletalscaffolding proteins (5). Anti-PDE4D antibodies are ideal tools for studying regulation,expression, phosphorylation, and protein-protein interactions of PDE4D variantsemploying pull-down immunoprecipitation protocol.
