Background Dapper homolog 1, an ortholog of Xenopus Dpr, antagonizes Wnt signaling by promoting Dvl degradation via a lysosome inhibitor-sensitive and proteasome inhibitor-insensitive mechanism. Dapper homolog 1 acts as an interacting protein for Dvl, and the interaction is formed between the DEP (Dishevelled, Egl-10, and pleckstrin) domain of Dvl and the central and C-terminal regions of Dapper homolog 1. The gene for human Dapper homolog 1 is localized on chromosomal region 14q23.1. The protein impedes the degradation of CTNNB1/beta-catenin, thereby enhancing the transcriptional activation of target genes of the Wnt signaling pathway. The protein is also required for normal notochord formation and is downregulated in hepatocellular carcinomas (HCC). It is expressed in human liver and various other tissues.
