Background NALP2 is a 1062 amino acid containing cytosolic protein, belonging to the NLRP family and is characterized by the presence of an N-terminal pyrin domain (PYD), eight leucine rich repeats and a NACHT domain. This protein is involved in the activation of caspase-1 by Toll- like receptors and may also be involved in protein complexes that activate proinflammatory caspases. The DAPIN domain of NALP2 helps in suppressing TNF- and CD40-induced NFKB1 activity at the level of the IKK complex, by inhibiting NFKBIA degradation induced by TNF. NALP2 is known to interact with CHUK, IKBKB and IKBKG, as well as with full-length PYCARD and with the DAPIN domain of NAPL1, but not the full-length protein. It is widely expressed in most of the tissues with high expression in lung, placenta and thymus, low expression in ovary, intestine and brain and variably in some cancer cell lines.
