Background NANOG is a unique variant of NK2 family of transcription factors. It is a human ortholog of mouse Nanog and consists of a highly conserved homeodomain and Trp-rich repeats. It is specifically expressed by embryonic stem cells and is crucial for second embryonic lineage determination. It works in concert with OCT-4 and SOX-2 and plays a central role in regulating pluripotency and self-renewal of ICM and ES cells. It functions independent of LIF/gp130/Stat3 pathway and prevents differentiation of ES cells to extraembryonic endoderm and trophectoderm lineages. Its expression has also been detected in embryonic carcinoma cells and germ line tumors indicating a therapeutic intervention.
