Background The E2 enzyme, Ubc13, and the E2 enzyme variants, Uevs, form stable, high affinity complexes for the assembly of Lys63-linked ubiquitin chains. This complex has been shown to activate NF-kB signaling pathway by linking TRAF6 to IKKs. Blockade of this polyubiquitin chain synthesis, but not inhibition of the proteasome, prevents the activation of IKK by TRAF6. These results suggest a new regulatory function for ubiquitin, in which IKK is activated through the assembly of Lys63-linked polyubiquitin chains. The UEV proteins are also involved in the control of differentiation.
