Background: The HIF1A gene encodes the HIF-1alpha subunit of hypoxia-inducible factor 1, a transcription factor that is essential for cardiovascular development and systemic O2 homeostasis (1). HIF-1 is found in mammalian cells cultured under reduced O2 tension and is necessary for transcriptional activation mediated by the erythropoietin gene enhancer in hypoxic cells. It has been shown that both HIF-1 subunits are basic-helix-loop-helix proteins containing a PAS domain, defined by its presence in the in the mammalian ARNT and AHR proteins. HIF-1 alpha is most closely related to Sim (2). HIF-1 binds to cis-acting hypoxia-response elements within the erythropoietin, vascular endothelial growth factor, and other genes to activate transcription in hypoxic cells. Results indicate that increased transcriptional activity mediated by HIF-1 in hypoxic cells results from both increased HIF-1alpha protein levels and increased activity of HIF-1alpha transactivation domains (3). |
