|
Background: Nck is a ubiquitously expressed adaptor protein consisting of one SH2 and three SH3 Src homology domains. Stimulation by epidermal growth factor (EGF) or platelet-derived growth factor (PDGF) leads to phosphorylation of Nck on tyrosine, serine and threonine residues. Overexpression of Nck resulted in transformation of NIH 3T3 and 3Y1 rat fibroblast (1-2). Additionally, Nck has been shown to associate with BLNK, PLCG1 and VAV1 in a B-cell antigen receptor-dependent fashion and may play a role in linking tyrosine and serine/threonine kinase pathways within the cell (2).
