Comments

Background: Nicastrin, a transmembrane glycoprotein, is an essential component of the gamma-secretase complex that is required for the intramembrane proteolysis of an increasing number of membrane proteins, including the amyloid-beta precursor protein (APP) and Notch (1). It probably represents a stabilizing cofactor required for the assembly of the gamma-secretase complex (2). Nicastrin regulates gamma-secretase cleavage of the amyloid precursor protein by forming complexes with presenilins, in which most mutations causing familial early-onset Alzheimer's disease (EOAD) have been found (3).
