Background: OCA-B is a lymphoid-specific transcriptional coactivator which interacts with the transcription factors Oct-1 and Oct-2 (1). It has been shown to be critical for development of a normal immune response, where it mediates octamer-dependent transcriptional activity in B lymphocytes. It is also critically involved in the formation of germinal centers in secondary lymphoid organs. OCA-B levels have been observed to be massively upregulated in germinal center B cells, as compared with resting B cells. SIAH1 and SIAH2, known regulators of protein stability, have been identified as two related OCA-B interacting proteins. SIAH1 has been suggested as part of a novel regulatory loop controlling the level of OCA-B protein in B cells. B-cell receptor cross-linking results in a degradation of OCA-B, which further results in a reduction of transcriptional activation of OCA-B-dependent reporters (2, 3). |
