Background: Rap1, also known as Krev-1 and smg p21, is a member of the Ras super family of GTPases closely related to Ras. Two known isoforms of Rap1 exists, Rap1A and Rap1B which share 95% identity (1-2). As a molecular switch, Rap1 cycles between two conformational states depending on whether GDP bound (active form) or GTP bound (inactive form). Rap1 associates with a subset of Ras effectors including Raf-1, B-Raf, phosphoinositide 3-kinase, Ral GEFs, and PLC (3). Although its function is still unclear, Rap1 has been shown to be involved in the regulation of integrin-mediated cell adhesion (4).
