Background SPHK1 catalyzes the phosphorylation of sphingosine to form sphingosine 1 phosphate (SPP), a lipid mediator which has both intra- and extracellular functions. Intracellularly, SPP regulates proliferation and survival. Extracellularly it functions as a ligand for EDG1. SPHK1 also acts on D erythro sphingosine and to a lesser extent sphinganine, but not other lipids, such as D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. Competitive inhibitors of SPHK block formation of SPP and selectively inhibit cellular proliferation induced by a variety of factors, including platelet derived growth factor and serum. SPHK1 has also been found to mediate TRAF2 activation (Xia et al 2002). TRAF2 is a member of the TNF receptor associated family and is required for TNF alpha activation of MAPK8/JNK and NFkB. SPHK1 is widely expressed, with highest levels in adult liver, kidney, heart and skeletal muscle.Synonyms: SPHK, SPK, SPK 1, SPK-1
