Background DNA replication in eukaryotes takes place in multiple sites of origins of replication scattered throughout the genome. The origin recognition complex (ORC) is a multiprotein assemblage originally identified in S. cerevisiae budding yeast that binds to the ARS (autonomously replicating sequence) consensus, a DNA motif which is an essential part of yeast origins of replication. The ORC complex consists of six distinct polypeptides (ORC1-ORC6), all having similar homology and essential for viability. ORC binding has also been implicated in transcriptional silencing at certain yeast loci. Our current understanding of the initiation of DNA replication in eukaryotes is based on studies done in yeast, but recent work has identified ORC proteins in higher eukaryotes. Mammals have close protein homologs to the yeast ORC proteins which are believed to be essential components of the mammalian origin recognition complex and may play a widespread role in packaging chromosomal domains through interactions with heterochromatin-organizing factors. There is a striking preferential but not exclusive association of ORC2 with heterochromatin on interphase and mitotic chromosomes. In Drosophila, ORC2 is highly concentrated at foci along the apical surface of the interphase nuclei, consistent with the known localization of pericentric heterochromatin. Consistent with this localization, ORC1, ORC2, ORC5 and ORC6 are found in a complex. Additionally, ORC2 has been shown to co-immunoprecipitate with ORC4. ORC2 has a molecular weight of approximately 72 kDa in western blot analysis. The antibody recognizes human ORC2. The N-terminal half of Hsorc2p fused to GST was used as immunogen.
