Background CrmA (cytokine response modifier A) is a cowpox virus-encoded serpin protein that inhibits apoptosis in a variety of systems. CrmA protects cells against apoptosis by inhibiting specific members of the ICE/CED-3 cysteine protease (designated caspase) family that are essential for apoptosis. One of the proteases inhibited by CrmA is caspase-3 (also known as CPP32, Yama, apopain). Caspase-3 is activated early in apoptosis and appears to be involved in the proteolysis of several important molecules. Activated caspase-3 is found only in cells undergoing apoptosis, and consists of 17 and 11 kDa subunits that are derived from a 32 kDa proenzyme (pro-caspase-3) by cleavage at multiple aspartic acid sites. Poly (ADP-ribose) polymerase (PARP) is a protein cleaved by activated caspase-3 during apoptosis. CrmA inhibits PARP cleavage, thereby blocking apoptosis. It is thought that if a particular apoptotic pathway can be inhibited by CrmA, then the activation of a caspase is necessary for apoptosis to take place. For example, CrmA has been shown to block Fas-mediated apoptosis, suggesting that activation of one or more caspases is required for Fas-induced cell death.
