Specificity This product was prepared from monospecific antiserum by delipidation and defibrination. Antiserum will specifically react with a 85 kDa cyclin T1 protein from human, rat and mouse tissue. No reaction was observed against other related cyclins. Cross reactivity with cyclin T1 from other species may also occur.

Application Details

Application Notes Recommended Dilutions: This product was assayed by immunoblot and found to be reactive against Cyclin T1 at a dilution of 1:500 to 1:1000 followed by reaction with Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] (Goat) code ABIN101990. Anti-Cyclin T1 is suitable for the detection by immunoblot of human, rat and mouse Cyclin T1. Suggested dilutions for ELISA are 1:500 to 1:2000. For immunohistochemistry on paraffin embedded tissue, dilute the product 1:200 to 1:1000. Optimal titers for other applications should be determined by the researcher. Note: Suitable for ELISA, immunoprecipitation, immunohistochemistry, immunoblotting and other immunological methods requiring high titer and specificity. The positive transcription elongation factor b (P-TEFb) is identified as cdk9 paired with cyclin T1. The cyclin T1/cdk9(PITALRE) complex phosphorylates the carboxy terminal domain of RNA Polymerase II in vitro and associate with HIV-1 generated Tat in HIV-1 infected T-cells. Tat stimulates human HIV-1 viral transcription elongation. This suggests that cyclin T1/cdk9(PITALRE) is one of the HIV-1 required host cellular cofactors generated during T cell activation. Cyclin T1/cdk9(PITALRE) is shown to interact with Tat to restore Tat activation in HeLa nuclear extracts depleted of P-TEFb. The cdk9(PITALRE) activity and cyclin T1 are essential for activation of transcription when tethered to the heterologous Rev response element RNA via the regulator of expression of virion Rev.
