Specificity This AE1/AE3 antibody cocktail recognizes most of the acidic (Type I) keratins as well as basic (Type II). It recognizes keratin polypeptides of 40, 48, 50, 52, 54, 56.5, 58, 59 and 64 to 67 kD. In keratinized epidermis, 50 kD keratin is present in the basal layer, while 56.5 kD keratin is present in suprabasal layers, where as 58 kD keratin is present in the basal and suprabasal layers, while 65 to 67 kD keratin is present in the cells above the basal layers. This antibody can be used to stain distinct cell layers in an epithelial tissue and various neoplasms of epithelial origin.

Application Details

Application Notes Titer/Working Dilution: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Enzymatic predigestion of paraffin embedded tissues is recommended prior to the immunostaining. Positive Control: Skin. Cellular localization: Cytoplasmic
