| Specificity | T cell activation through the antigen receptor (TCR) involves the cytoplasmic tails of the CD3 subunits CD3., CD3d, CD3e and CD3. (1, 2). These CD3 subunits are structurally related members of the immunoglobulins super family encoded by closely linked genes on human chromosome 11 (1). The CD3 components have long cytoplasmic tails that associate with cytoplasmic signal transduction molecules. This association is mediated at least in part by a double tyrosine-based motif present in a single copy in the CD3 subunits. CD3e may play a role in TCR-induced growth arrest, cell survival and proliferation (3). This epitope affinity purified rabbit polyclonal antibody is specific for CD3. |
Application Details: CD3 (Human) antibody
| Application | Immunohistochemistry (Paraffin-embedded Sections) (IHC (p)), Enzyme Immunoassay (EIA), Western Blotting (WB) |
| Application Notes | ELISA: 0.1-1.0 µg/ml WB: 0.5 - 2 µg/ml IHC: 1:100 for 10 min at RT By IHC on human tonsil, a postive cell membrane staining is observed in Formalin-fixed tissues (boiling in 10 mM citrate buffer, pH 6.0 for 10 min, followed by cooling at RT for 20 min). Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user: The results might be affected by several factors, including secondary antibody affinity, antigen concentration, sensitivity of detection method, temperature and length of incubations. |
| Concentration | 0.5 mg/ml |
| Buffer | This polyclonal antibody is supplied as an epitope affinity purified rabbit IgG, 100 µg in 200 µl of phosphate buffered saline (pH 7.4) containing 0.02% sodium azide. |
| Storage | Store at 2-8 oC for up to one month. Store at –20 oC for long-term storage. Avoid repeated freezing and thawing. Shelf life: one year from despatch |
