| Specificity | Recognises a different epitope than clone 4H9 (ABIN111808). 5D5 mAb binding decreases only partly after CA treatment. It is possible that the antigen determinant is exposed more openly after DEP modification of histidine residues, whereas this determinant becomes barely accesible after lysine modification. Human serum, containing CRF-binding protein, inhibits the binding of CRF to this antibody. Reacts with CRF. Does not react with bovine serum albumin, insulin, ovalbumin or epidermal growth factor. Expected to react with rat CRF, although not tested. Dissociation constant: Averages at 1.5 x 10(e)-8 M for native CRF, and 2 x 10(e)-9 M for modified CRF. |

| Application Notes | ELISA. High binding constant allows this antibody to be used in a sandwich ELISA with clone number 4H9 (ABIN111808). Sensitivity: 25 pg/ml. Immunohistochemistry on paraffin embedded sections: A 4 ug/ml solution has worked well on paraffin sections, after microwave pre-treatment. 0.1% trypsin digestion may also be necessary. The detection system used was avidin-biotin. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user. |
