| Description | Cyclin A is a 60 kDa cyclin that binds both cdk2 and p34cdc2 (cdk1), giving two distinct cyclin A kinase activities, one appearing in G1/S phase, the other in G2/M. The level of cyclin A protein peaks in S-phase, and the activity of the cdk2-cyclin A complexes is essential for DNA replication and progression through S phase. Cyclin A is thus a marker for actively proliferating cells and for cells in S-phase. In normal cells, cdk2-cyclin A complexes associate with PCNA and p21Cip1/WAF1. In addition, cyclin A has been implicated in cell transformation and is found in complexes with E1A, transcription factors DRT F1 and E2F, and retinoblastoma protein, p110. |

| Application Notes | SDS-PAGE & Western Blotting 1) Wash the cells 3 times with PBS and suspend with 10 volumes of cold Lysis buffer (50 mM Tris-HCl, pH 7.2, 250 mM NaCl, 0.1% NP-40, 2 mM EDTA, 10% glycerol) containing appropriate protease inhibitors. Incubate it at Western blot analysis of Cyclin A expression in Jurkat cells (1), Raji cells (2), HeLa cells (3), WR19L cells (4) and PC12 cells (5) using Western blotting: 1 µg / mL for chemiluminescence detection system. Immunoprecipitation: Not recommended. Immunohistochemistry: Not tested. Immunocytochemistry: Not recommended. Flow Cytometry: Not tested. |
