| Description | DNA ligases are ATP-dependent, nuclear enzymes that catalyze the joining of single and double-strand DNA breaks. Four distinct DNA ligases, designated DNA Ligase I-IV, have been identified in mammalian cells. The 125 kDa DNA Ligase I is a phosphoprotein responsible for joining Okazaki fragments during lagging strand DNA replication. In cooperation with replication factors A and C, DNA polymerase epsilon, and PCNA, it also plays a role in the base and nucleotide excision repair pathways. Deficiencies in DNA Ligase I result in hypersensitivity to DNA-damaging agents and aberrant DNA repair and replication. |
