| Purification | Prepared by immunizing donkeys with the above antigen. The resulting antiserum is then delipidated and, if necessary, adsorbed to monospecificity by use of solid-phase adsorbents. Following adsorption, the antiserum is affinity purified by binding to the specific antigen attached to an inert matrix and the bound immunoglobulin is then eluted with chaotropic buffers. The immunoglobulin is then conjugated with fluorescein isothiocyanate (FITC) and unrelated fluorochrome is isolated by gel filtration. |
