| Purification | This antibody was produced by antigen dependent affinity chromatography of positive sera subsequently to immunization of NZW rabbits with Cys–(Histidine)14 coupled to KLH. The purity of antibody fractions was determined by Coomassie staining upon SDS-PAGE. Subsequently the antibody was conjugated to HRP by periodate coupling. Functional analysis of the purified antibody revealed strong and specific reactivity against recombinant HIS-tag proteins by Western Blotting, ELISA and Flow Cytometry. Hereby it makes no difference if the HIS-tag of the target protein is fused to the N- or the C-terminus. |

Determination of a bacterially expressed recombinant proteins with AntiHIS-tag-HRP by Western Blot. Protein extracts of E.coli strains expressing 3 individual recombinant proteins of 38kD, 50kD and 68kd (from right to the left) were resolved by SDS-PAGE and subsequently transferred to PVDF membrane. The proteins were determined by AntiHis-tag-HRP (1µg/ml) in combination with colorimetric visualization. The numbers indiacted on the right represent the molecular weight of the protein standard used in the experiment. |
