| Description | Haptoglobin (Hp) is a plasma protein mostly synthesized in the liver. The haptoglobin monomer consists of two heavy, beta-chains (40 kDa) and two light, alpha-chains (alpha-1, 9 kDa, alpha-2, 16kDa) linked by disulphide bonds. Hp is a heterogeneous protein, the major variation being determined by two alleles at the alpha locus, namely alpha-1 and alpha-2. These associate to form three major Hp types, Hp1-1, Hp1-2 and Hp2-2. Hp1-1 exists as a monomer (alpha-1-beta) 2 (98kDa), Hp1-2 is polymeric (alpha-1-beta) m. (alpha-2-beta) n (about 200 kDa), as is Hp2-2 (alpha-2-beta) n (about 400 kDa). Plasma haptoglobin concentrations range from 1-3g/L. Hp has a high affinity for hemoglobin (Hb) and its function appears to be to prevent loss of Hb in urine, which would lead to loss of iron. The Hp Hb complex is catabolized in the liver. Hp levels are decreased in hemolytic anemias and increased in a variety of inflammatory conditions. |
