| Description | This antibody is a useful tool to study molecular mechanisms associated with the G2 toM transition and chromatin condensation and for the screening of in vivo inhibitors ofkinase(s) or phosphatase(s) involved in H3 phosphorylation ordephosphorylation.Mitogenic stimulation, oncogen transformation, orinduction ofoncogenic ras expression is accompanied with increases in Ser-10 phosphorylation ofthe N-terminal domain H3 (ref 5).Indeed, it has been shown that phosphorylated H3 isassociated with c-fos and c-myc genes in stimulated cells (ref 6).H3 phosphorylationmay contribute to proto-oncogen induction by modulating chromatin structure andreleasing blocks in elongation.Site-specific phosphorylation of histone H3 at serine 10(Ser10) appears to occur exclusively during mitosis in mammalian cells. H3dephosphorylation occurs quite rapidly after mitosis and Ser10 remainsunphosphorylated throughout the remainder of interphase (See Ajiro et al. and Wolffe). |
