| Specificity | Phosphorylation and dephosphorylation of cellular proteins are central steps in transducing extracellular signals to the cell nucleus. Phosphorylated epitopes may serve as docking sites for the assembly of protein complexes or may alter the 3-dimensional protein structure thus modulating enzymatic activity or the ability to undergo protein-protein-interactions. Modification of proteins on tyrosine residues is mediated by protein tyrosine kinases. Tyrosine phosphorylation may alter the biological activity or mediate the assembly of protein complexes via interaction of phosphotyrosine residues with SH2 or PID domains. ABIN126875 recognizes phosphotyrosine in the context of the surrounding amino acids, showing high preference for phosphorylated EGF receptor. It reacts with human, mouse, rat and dog. |

| Application Notes | Suitable for Immunoprecipitation (1 - 10 µg per 106 pervanadate-treated A431 cells, use phosphotyrosine MW standardas positive control), Western Blot (0.5 µg/ml for HRPO/ECL detection, recommended blocking buffer CPPT: 0.5% (w/v) casein, 1% (w/v) PEG 4000, 1% (w/v) Polyvinylpyrrolidone (PVP), 0.1% (v/v) Tween 20, 2x PBS) and ELISA (0.05 µg/ml). Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user. |
