Western Blotting (1:1K-5K for neat serum and 1-10 µg/ml for affinity pure using Chemiluminescence technique). An antibody raised against a similar peptide has been shown to react with a SERT in baculovirus cells expressing recombinant rat and human-SERT (2). Antibody activity may be reduced considerably when detecting endogenous SERT from rat or human cells. SERT has been observed to migrate as 94 kDa (rat platelet), 80 kDa (pulmonary membranes, and 76 kDa (rat brain) (2). This could be due to differences in postranslational modifications (e.g., glycosylation). ELISA (1:100K, using 50-100 ng SERT control peptide/well). Immunoprecipitations . An antibody to the SERT epitope has been shown to be effective in immunoprecipitating the 61 kDa species from rat and human SERT-transfected HeLa cells (2). Histochemistry & Immunofluorescence: Not tested. We recommend the testing of affinity purified antibody at 10-50 µg/ml on frozen sections of tissues fixed with 4% paraformaldehyde.
