Specificity The antibody recognizes the truncated version of HER 2, HER-2 ECD, which has a wide tissue distribution similar to that of EGF-R. The epitope recognised is a perinuclear cytoplasmic protein. Does not react with EGF-R. The HER-2 oncogene is located on the long arm of chromosome 17 at 17q21 and encodes a 185 kDa transmembrane tyrosine kinase receptor belonging to the EGFR family. The HER-2 oncogene is overexpressed in about 30% of human breast and ovary carcinomas, where overexpression correlates with a more aggressive clinical course.

Application Immunohistochemistry (Frozen Sections) (IHC (fro)), Immunofluorescence (IF), Immunoprecipitation (IP), Immunohistochemistry (Paraffin-embedded Sections) (IHC (p)), Western Blotting (WB)
Application Notes Immunofluorescence (has given very good results when tested on breast cancer tissue samples in indirect IF). Immunoprecipitation. Immuno-histochemistry on frozen sections (Will work on formalin fixed, paraffin embedded tissue sections from patients where HER 2 is over expressed, eg. breast, stomach, ovarian and bladder carcinoma). Western blotting. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user.
Concentration 0.77 mg/ml (OD280, lot specific)
Buffer This antibody is supplied as liquid, purified immunoglobulin fraction (prepared by caprilic acid precipitation) in PBS buffer pH 7.2 with 10 mM sodium azide as preservative.
Storage Aliquot and store the antibody at –20°C. Avoid repeated freezing and thawing. Shelf life: one year from despatch.
